This happens after returning home to a surprise birthday party for the real Big Momma. His discovery is witnessed by Sherry, who tells him she had no idea what Lester was planning, and did not report her key was stolen out of fear of being fired. Two undercover FBI agents, Malcolm Turner and John Maxwell, find themselves assigned by their boss to track down the escaped convict, Lester Vesco, who was serving a life sentence for murder and the armed robbery of a bank. Malcolm and John learn that Lester may be seeking to find his ex-girlfriend, Sherry Pierce, an employee of the bank who is suspected of supplying him with a key to the vault, as the money that was stolen was never recovered. Discovering that Bishop plans to kill Tom as part of the deal he made with his customer, Malcolm saves his life and ensures Bishop cannot escape before the FBI arrives. Although shocked to discover Malcolm's real identity, Tom is thankful to him when he informs Morales that he was being coerced by his boss under the threat of having his family killed for refusing.

Discovering Morales is sending an agent undercover to secure the job of nanny within Tom's family, Malcolm decides to take advantage of this, lying to Sherry that he must go out-of-state for a conference. Matters soon become complicated when John discovers that the real Big Momma has returned home. In his efforts to stop her re-entering the house, John instructs Nolan to lock her out, only for the dim-witted guard to accidentally lock Malcolm out. Malcolm swiftly breaks into the house, whereupon he tackles Lester and subdues him after John is shot, but at the cost of his disguise being dropped in front of everyone, including Sherry and Trent. After Lester is arrested and the money recovered, Malcolm finds that Sherry and Trent refuse to speak to him, heartbroken by his deception. When a street-smart FBI agent is sent to Georgia to protect a beautiful single mother and her son from an escaped convict, he is forced to impersonate a crass Southern granny known as Big Momma in order to remain incognito.

FBI agent, Malcolm Turner and his partner, John are assigned to capture a escaped convict known as Lester Vesco, who was originally serving a life sentence in prison but escaped his cell and killed a prison doctor and took off in his car. Malcolm and John are sent to Cartersville, Georgia to stake out the house of a elderly woman, Hattie Mae Pierce (simply known as "Big Momma") who is the grandmother of Lester's ex-girlfriend, Sherry Pierce. The FBI suspects that Sherry aided him during the robbery by giving him the key to the vault. Big Momma unexpectedly leaves town for a couple of weeks, the two sneak into her house and Sherry calls the house and Malcolm disguises his voice as Big Momma in order to lure Sherry and possibly obtain a confession. Malcolm then disguises himself as Big Momma until he can get a confession from Sherry and nab Lester. Malcolm and John stake out the house of Sherry's estranged grandmother, Hattie Mae Pierce—an obese, elderly African American woman, whom her friends affectionately call Big Momma—in Cartersville, Georgia.
Big Momma forgives him for his actions, and the church crowd cheers as Malcolm and Sherry kiss, before Big Momma and the choir sing "Oh Happy Day" during the film's closing credits.
